Conclusion Our results illustrate that differences in the normalized fractional abundance of fragment ions in top-down mass spectra are correlated with subtle changes in the tertiary protein structure and charge localization. Using quantitative statistics, small but significant perturbations in protein structure can be detected, and the method allows for comparison between any two conditions of interest.
