A molecular replacement search with the kcTRP5b.1 model yielded a highly significant solution; subsequent refinement and iterative model building revealed good agreement between the design and crystal structure across the core alpha helices (Fig. 6a ), but divergence in two of the four repeat-repeat loop connections (Fig. 6b and Supplementary Fig. 10a ) leading to a globally dissimilar topology.
