Polygenic risk affects the progression from A+T− to A+T+ The survival analysis showed a marginally significant contribution by APOE-e4 burden (HR = 1.62; 95% CI: 1.05–2.51; P = 0.031), which was mainly driven by APOE-e4 homozygotes (HR = 2.58; 95% CI: 1.05–6.35; P = 0.039) rather than APOE-e4 heterozygotes (HR = 1.74; 95% CI: 0.87–3.49; P = 0.12) ( Fig. 2 ).
