In reality, all tumor deconvolution tools may experience a decrease in deconvolution performance as the tumor fraction increases, possibly due to several reasons: (1) as the tumor progresses to later stages, the intra-tumor heterogeneity may increase, and (2) in the later stages of tumor development, there may be significant changes in the components of cfDNA due to the dynamic evolution of the tumor microenvironment, etc. 28 These phenomena could affect the performance of tumor deconvolution tools for samples from the middle and late stages of tumor development.
