One important measure of the reliability of any screen is to confirm that most of the positive control sgRNAs (essential genes) and negative control sgRNAs (scramble and/or non-targeting) behave as they are expected to—displaying highly significant drop-out and non-significant changes, respectively ( Figure 9 A).
