Patients with BRAF mutation had a trend toward a worse overall survival with the addition of panitumumab (HR = 1.4; [0.82–2.39]). 45 On the other hand, a combined analysis of the CRYSTAL and OPUS studies showed a nonsignificant trend in RR, progression-free survival (PFS), and overall survival (OS) with the addition of cetuximab to FOLFIRI in BRAF mutant patients. 46 While these data are limited by the small portion of patients with BRAF mutation in each of the above studies, they suggest a lack of a clinically meaningful improvement in OS with the integration of anti-EGFR therapy with chemotherapy or as monotherapy in mCRC with BRAF mutations.
