Effect of orforglipron on body weight reduction The pooled meta-analysis of four studies demonstrated a highly significant reduction in body weight with Orforglipron compared to placebo (MD: -6.08%; 95% CI, -7.68 to -4.47; P < 0.01) (Fig. 1 ).
A subgroup analysis across five dosing regimens (3 mg to 45 mg) suggested a trend toward differential effects across doses ( P = 0.44, I 2 = 0%) (Figure S12 ), Although a numerical trend toward greater VLDL cholesterol reduction with higher doses was observed, the absence of statistical significance suggests that a clear dose-response relationship could not be established within the range of doses evaluated.
