Although a 10-fold decrease may be significant in terms of expression of some genes, in repeated attempts, the fold changes at the mRNA level differed with the batch of base medium used, so no definitive in vitro culture conditions that reliably mimic the midgut of the unfed tick were identified with regard to P66 production.
