Although it has been suggested that the search for interactions should be informed by biologic plausibility ( 11 ), experience from the study of genetic main effects, where hypothesis-generating discovery approaches revolutionized the field ( 2 ), suggests that such an approach, not limited to those SNPs with extremely significant main effects, may be valuable in detecting gene–lifestyle interactions.
