Though there was a nominal SNP × metformin interaction in the fully adjusted model ( P = 0.04), the observed trend was in the opposite direction from the expected prevention effect; the C allele conferred no detectable advantage on metformin recipients in diabetes prevention but was associated with a nonsignificant trend toward increased risk of diabetes (HR per copy of the C allele 1.17 [0.96–1.42], P = 0.13).
