These studies indicate that the YopN CBD mutants defective in the regulation of Yop secretion, with the exception of YopN(G44-Q47A) and YopN(S48-D51A), still bind the SycN/YscB chaperone; however, it is possible that a subset of these mutants exhibit subtle, but possibly significant, alterations in SycN/YscB chaperone binding that cannot be differentiated via DSP cross-linking and immunoprecipitation.
