Interestingly, we found a highly significant ( P < 0.001) increase in nuclear CUG-foci frequency in DM1 fibroblasts derived from two different patients (DM1 and DM1′) upon knockdown of DDX6 (foci elevated from 4.4 ± 0.28 and 7.9 ± 0.34 foci per cell respectively, to 7.1 ± 0.48 and 9.7 ± 0.37 per cell, respectively) (Figure 2A and quantified in Figure 2D ).
