Correlation of UCE Capture with Specimen Age and DNA Quality We found that all investigated variables (pre-library DNA concentration, post-library DNA concentration, raw read count, UCE locus count, UCE mean contig length and UCE reads on target) decreased with increasing specimen age, and this negative correlation was highly significant in all cases (R = -0.36–0.75 and p < 0.001; Table 2 , Fig 2A–2E ).
