The cell spreading area quantified from the F-actin staining showed an increasing trend as the substrate stiffness increased (Fig. 1D ), in that the average cell spread area was <1000 μm 2 on the softest micropillars (0.012 nN/μm) but >2000 μm 2 on the stiffest micropillars (1.014 nN/μm).
