Using the CRISPRater algorithm to predict efficient sgRNAs in a discrete manner yielded a highly significant selection of efficiently-acting sgRNAs—improving library design by 1.27-fold while retaining 91.2% of the libraries’ sgRNAs (combined medium and high efficiency groups), or by 1.19-fold while retaining 24.0% of the sgRNAs (high efficiency group only) (Figure 5A ).
