The study was not powered to demonstrate an EFS benefit, but an exploratory analysis at 5-year follow up showed a numerical trend in favor of the docetaxel/trastuzumab/pertuzumab arm compared with docetaxel/trastuzumab (84% versus 81%; HR 0.69; 95% CI 0.34–1.40). 40 TRYPHAENA was a three-arm study enrolling 225 patients with HER2+ tumors 2 cm or larger. 39 It primarily aimed to evaluate the cardiac safety of three pertuzumab and trastuzumab-containing regimens.
