Bacteria are freely moving inside the GUV, and temporarily overlapping cells (in z -direction) as well as out-of-focus cells are difficult to distinguish by the automated image analysis. Nevertheless, we clearly observed an increasing trend with about 18 cells after the 10 h cultivation, corresponding to approximately four generations.
