In a prespecified secondary analysis of 25,982 patients (94%) in FOURIER who had an LDL-C measured at 4 weeks (and who had not experienced a primary end point event), there was a highly significant linear relationship between lower LDL-C concentrations and lower risk of the primary and secondary efficacy composite end points; this extended to the bottom first percentile (LDL-C < 0.2 mmol/L). 39 Conversely, no significant association was observed between achieved LDL-C and safety outcomes, either for all serious adverse events or any of the other 9 prespecified safety events.
