To provide a hypothesis of functional significance of the polymorphisms found in the context of CGRP signalling and migraine, it is mandatory to consider that the data that have resulted close to significance but without reaching it may have been affected by: (1) the small sample size of the cohorts, underpowered to detect small genetic main effects and for further stratification analysis; (2) the confounding influence of environmental factors.
