Therefore, we assume that net negative acidic peptides (net charge < 0) were unfavorable under our RNA-binding condition, resulting in a constrained chemical parameter space of the enriched peptides after seven rounds of selection. A clear trend toward decreasing hydrophobicity in both libraries may indicate that the selected peptides do not prefer forming a tertiary soluble structure with hydrophobic core, in order to maximize the interface with the unstructured linear RNA.
