Indeed, although a phase II trial investigating carboplatin, paclitaxel, and vorinostat in advanced NSCLC patients showed a trend toward improved ORR with the addition of vorinostat ( NCT 00481078 ) [ 21 ], the subsequent phase III trial in the same population failed to meet the primary endpoint of overall survival (OS) in a pre-specified interim analysis, revealing a negative impact of vorinostat on clinical outcomes ( NCT00473889 ) [ 22 ].
