While MEKiAUTO ( NCT04214418 ) showed the combination of mitogen-activated protein kinase kinase inhibitor (MEKi) cobimetinib, HCQ, and PD-L1 inhibitor Atezolizumab demonstrated limited tolerability and efficacy in patients with advanced KRAS-mutated pancreatic ductal adenocarcinoma, clinical benefit was observed in 36% of patients, while all of whom achieved stable disease as their best response; and patients with KRAS G12R mutations showed a trend toward improved overall survival (OS) compared to those with KRAS non-G12R mutations ( 77 ).
