While differences in PFS and response rates compared to fulvestrant did not reach statistical significance, lasofoxifene was associated with a more pronounced reduction in circulating ESR1 mutant allele fraction (82.9% drop from baseline at week 8 compared to 61.5% in the fulvestrant arm), suggesting potential biological activity warranting further investigation ( 44 ).
