47 , 48 , 50 , 51 In uterine fibroids specifically, cross sectional and case-control work report inverse associations between serum vitamin D and fibroid prevalence or burden, and one randomized clinical trial using weekly 50,000 IU for 12 weeks suggests that supplementation inhibited fibroid growth compared with a placebo group that experienced fibroid enlargement over the same period, although absolute shrinkage in the intervention arm did not reach statistical significance within that short timeframe. 51 , 52 These human data align with robust preclinical evidence that vitamin D analogs reduced fibroid cell proliferation and fibrotic marker expression in vitro and in animal models, supporting a plausible antifibrotic effect that supports the need for longer, larger human trials. 53 , 54 Adenomyomas, though less extensively studied, show a similar pattern.
