However, posterior wall thickness at diastole (PWTD) was significantly higher in KO hearts ( Figure 10 A ). Furthermore, PKN1 KO hearts had a trend towards altered functional characteristics compared to wild type hearts under conditions of decreased preload (IVC occlusion), including decreased end systolic pressure-volume relationship (ESPVR), increased end diastolic pressure-volume relationship (EDPVR) and decreased preload recruitable stroke work (PRSW), although these changes did not reach statistical significance (see Supplementary material online, Figure S8 ).
