Although the accessible and polar surface areas showed a trend in an improved affinity ( Table 1 ) for Aβ fibrils ( 4 vs 5 : K i = 353 vs 91 nM; 7 vs 8 : K i = >500 vs 505 nM) and a reduced affinity for Asyn fibrils ( 4 vs 5 : K i = 531 vs 906 nM; 7 vs 8 : K i = 53 vs 96 nM) to the compounds with the methoxy group versus dimethylamino group, the tendency did not consistently show in the nitro-containing compounds.
