Although the association of the Q152E variant with SLE was only marginally significant ( 81 ), of interest is that the Q152E substitution is located within the CD22 extracellular domain (at the interface between Ig domains 2 and 3) and introduces a charge difference; since it is located far from the SA-binding pocket, it is unlikely to directly affect CD22 binding to α2-6 sialylated ligands; however, this polymorphism might affect other aspects of CD22 biology such as stability, adhesion and trafficking.
