Western analysis of hippocampal K48-linked polyubiquitinated proteins, generally targeted for proteasomal degradation, showed a clear trend of increased levels in PA28αOE (Fig. 4 b and Additional file 13 ; P = 0.051), indicating reduced 26S proteasome activity and in line with PA28α overexpression being unable to activate proteasome activity. 20S proteasome capacity was not induced in PA28αOE hippocampi (Fig. 4 c and Additional file 10 ) and hippocampal levels of proteasome related markers Rpt2 (19S subunit), β5 (20S) and β5i (20Si) did not differ between WT and PA28αOE (Fig. 4 d and Additional file 13 ), while PA28α overexpression in PA28αOE hippocampi was verified (Fig. 4 d).
