ΔΔ G fold analysis of substitutions in the StEP library indicated a clear trend that ΔΔ G fold is a predictor for the recombination experiments after analysis of active and inactive variants. “Intra‐” and “inter‐category” recombinations by site‐directed mutagenesis were performed in a stepwise manner as in previous reports. 31 , 32 Based on the obtained results of, in total, 84 recombinants (Figure 2 and Table S3 in Supporting Information) thresholds for recombining beneficial substitutions are postulated as the CompassR rule in the Results section.
