A truncating insertion to amino acid 266 was chosen as the representative ALE mutation to pykF for the toxic isobutyric acid condition since it represents a clear trend in disrupting the PykF subunit interfaces and manifested in two independent ALE replicates with this selection pressure ( Figure 1 workflow step 5.3).
