Compared to previously reported colorimetric assay-based approaches 29 – 32 , utilizing growth as a simple readout afforded higher throughput which enabled us to observe a strong trend of convergent evolution: multiple NMN + -reducing variants employ very different sets of mutations to achieve the same predicted mechanism (recapitulating AMP) to enhance the NMN + -dependent activity.
