Although, these data did not reach statistical significance, there appeared to be a clear trend suggesting that the POLD1 R689W variant functionally impaired the stability of DNA fibers, as shown by the decreased elongation rate upon IR, while having no effect on the ongoing replication process despite replicational stress, as demonstrated by virtually unchanged second pulse origins and stalled replication forks specifically in POLD1 R689W/- cells.
