When we compared average read count to fold change in poly(A) tail lengths between 4SA and 4SD cells, the number of genes with significantly longer tails increased with read count ( Supplementary Figure S6C ), highlighting the possibility that the observed difference in poly(A) tail length is indeed transcriptome-wide, but highly expressed genes are more likely to be significant in our sequencing-based method.
