SNPs that remained nominally significant (p = 0.03) and therefore contributed to explaining additional THP variance after adjusting for rs12917707 were rs115312009, rs147825435, rs76619864, and rs75432530.
However, multiple known associations with TSH levels in European ancestry were nominally significant in African Americans, including PDE8B (rs2046045 p = 0.03, β = −0.09), VEGFA (rs11755845 p = 0.01, β = −0.13), and NFIA (rs334699 p = 1.50×10 −3 , β = −0.17).
These associations were also identified as nominally significant in CTRP, whereby GO process “Growth” is associated with PC2 ( P = 0.03) and “Regulation of Apoptosis” is associated with PC3 ( P = 0.01) and PC5 ( P = 0.01).
We also found nominally significant discrimination of unaffected siblings from healthy controls based on successful stop activation maps (AUC = 0.59, p < 0.030, 95% Modified Wald CI: 0.52–0.66 AUC) as well as nominally significant discrimination between participants with ADHD and healthy controls based on failed stop activation maps (AUC = 0.60, p < 0.019, 95% Modified Wald CI: 0.54–0.66 AUC, Fig. 1 ; for balanced accuracy, sensitivity and specificity measures in the complete and matched sample see Supplementary Table 2 ).
There was a nominally significant excess of heart failure (HR 1.34, 95% CI:1.03 to 1.75, p=0.03) but no associations with other studied cardiac outcomes.

Among all methods and metabolites, only the weighted median estimate for GCST90277281 indicated a nominally significant association (OR = 0.93, 95% CI: 0.87–0.99, P = .031), suggesting a possible weak inverse effect.
